The effects of various conc of (10-11, 10-9 and 10-7 M) 24-epibrassinolide (24-epiBL) on polyamines and auxins contents was analyzed in 7days old seedlings of Raphanus sativus L. cv. Pusa chetki under copper metal stress. Cu metal (0.2mM) treatment significantly increased endogenous levels of putrescine, spermidine and spermine when compared with control values. Cadaverine content got decreased under metal stress. Endogenous polyamines showed varying levels in seedlings when applying 24-epiBL treatment only. Maximum increase was found in putrescine, cadaverine and spermidine in 10-11, 10-9 and 10-7 M conc of 24-epiBL respectively when applied alone to R. sativus seedlings. However, metal treatment supplemented with 24-epiBL revealed small increase in... |

Cobalt (Co) affords both beneficial as well as toxic effects to plants. The present study was performed with an aim to find out the varietal differences among five tomato cultivars against the Co induced changes in growth, photosynthesis, nitrate reductase (E.C.1.6.6.1), carbonic anhydrase (E.C.4.2.1.1), antioxidative enzymes i.e. peroxidase (E.C.1.11.1.7), catalase (E.C.1.11.1.6), superoxide dismutase (E.C.1.15.1.1) and that of proline content. Seeds of tomato (varieties, K-25, NTS-9, NBR-Uday, Sarvodya, and Malti) were soaked in 0, 100, 200 or 300 µM CoCl2 for 0, 4, 8, 12 h (shotgun approach) and sampled at 30 days after sowing. All the varieties showed significantly different response to different treatment combinations. Despite substantial varietal... |

The phytohormone ethylene is involved in several physiological and developmental processes in higher plants, including ripening of fruits, abscission of organs and tissues, senescence, wound response as well as in other abiotic stresses. The enzyme 1-aminocyclopropane-1-carboxylate oxidase (ACO) catalyzes the last step of ethylene biosynthesis. The production of ethylene and the expression of a Coffea arabica ACO gene during the last stages of fruit maturation were investigated. A rapid increase of ethylene production at the green-yellow stage of fruit ripening, after the end of endosperm formation, and a decrease of ethylene production at the cherry stage indicates a climacteric phase during ripening. An ACC oxidase (Ca-ACO) from coffee fruit cDNA was... |

In the cerrado region of Brazil, no-tillage is an alternative management practice to conserve soil and reduce cost of crop production. However, many studies have been showed that there is a decrease in the yield when upland rice is cropped in this system. Therefore, there is a need that new breeding initiatives introgress favorable traits into upland rice for no-tillage adaptation. Key traits include faster emergence, ability to germinate when deep seeded, faster root development, and early canopy closure during the vegetative stages. All those traits can be summarized as "early vigor". So, the objective of this study was to evaluate physiological traits of four contrasting parental upland rice cultivars to verify their adaptation to no-tillage system. A... |

Cratylia floribunda seeds were ground and the clean crude saline extract was fractionated into albumin, globulin, prolamin, acidic and basic glutelin protein fractions. These protein fractions were examined for the presence of an endogenous lectin receptor by SDS-polyacrylamide gel electrophoresis, western blot, affinity chromatography on a Sepharose 4B-Cratylia floribunda (CFL) lectin column and kinetic analysis in real time by surface plasmon resonance (SPR). Prolamin was the richest protein fraction although very poor in haemagglutinating activity. Basic glutelin was far the less interesting fraction for lectin activity and protein content, even though this fraction contains considerable amounts of carbohydrates. Lectin was present in all protein... |

During ripening many important physic-chemical changes contribute to fruit quality, and they are precisely determined by gene expression. Specific genes are essential to normal ripening; however, information on gene expression about the majority of tropical fruit, such as bananas and mangoes is limited. In this way, the present study was undertaken with the objective to provide preliminary access to the changes in expression of some genes potentially relevant to banana and mango ripening. To simultaneously evaluate the changes in gene expression, a small collection of genes related to ethylene biosynthesis, starch mobilization, cell wall disassembly, pigment synthesis and ascorbate metabolism was assembled in nylon membranes and probed with cDNA from... |

Tolerance to drought in plants is not a simple trait, but a complex of mechanisms working in combination to avoid or to resist water deficit. Genotypes that differ in tolerance to water deficit may show qualitative and quantitative differences in gene expression when submitted to drought periods. Four cotton (Gossypium hirsutum L.) genotypes (Siokra L-23, Stoneville 506, CS 50 and T-1521) with contrasting responses to water deficit stress were studied using the Differential Display (DD) technique to identify and isolate genes which may differ among them. Fifty-two cDNA fragments differentially expressed during water deficit were isolated, cloned and sequenced. Search of gene bank databases showed that two cDNA clones, A12B15-6 and A12B13-1, have high... |

Starting from semihardwood branches of peach and plum rootstocks (Prunus cerasifera E.) cv. MRS2/5, plum rootstocks (Prunus cerasifera × P. salicina) × (P. cerasifera × P. persica) cv. Ishtarà and olive tree (Olea europaea L.) cv. Frantoio, cuttings were collected and treated with 0 mg.L-1 and 2500 mg.L-1 of indolebutyric acid (IBA) by immersion in the base of the cuttings. After 60 days of planting, the cultivars MRS 2/5, Ishtarà and Frantoio presented a significative difference with relation to the rooting induction by application of the indolebutyric acid (27.5, 47.5 and 41.25 %, respectively), inducing a higher number of roots per cutting without at the same time significantly increasing the length of the roots themselves. The levels of ascorbate and... |

A greenhouse experiment was conducted to investigate the effect of flooding on two tomato cultivars and two wild related species. Forty days old tomato plants were subjected to a continuous flooding stress of different durations: 0, 2, 4 and 8 days. Plant pots were placed inside larger plastic pots; they were irrigated with excessive quantity of tap water at 25ºC so that the level of water above the surface of soil was 15 cm throughout the flooding period. At the bottom of each plastic pot a drilled hole allowed complete drainage of the pot after flooding. Parameters studied include plant height, number of leaves, leaf length, chlorophyll fluorescence, chlorophyll content, wilting, leaf senescence, adventitious root formation, number of flowers and fruits... |

This work focuses on the evaluation of flowering and vegetative growth of olive tree submitted to pruning and paclobutrazol application under field conditions with low temperatures during the winter. Two-years-old olive plant variety Grappolo 575, were submitted to light pruning, removing apical dominance, before treatments application. The treatments were organized in a 4×2 factorial scheme, respective to four PBZ concentrations tested: 0, 200, 400 and 800 mg L-1 of PBZ and two plants groups, with and without pruning, in randomized block with four replications. Paclobutrazol concentrations tested did not affect the olive tree flowering. The plant vegetative growth was reduced until 60 days after paclobutrazol application. Pruning resulted in stimulation... |

Glutamate metabolism and amino acid translocation were investigated in the control and cadmium stressed shoots of tomato (Solanum lycopersicum - 63/5 F1), using (15N) ammonium and (15N) glutamate tracers. Regardless of organ type, [15N] ammonium assimilation occurred via glutamine synthetase (EC 6.3.1.2), in the control and stressed plants, and it did not depend on glutamate dehydrogenase (EC 1.4.1.2). The [15N] ammonium and ammonium accumulation patterns support the role of glutamate dehydrogenase in the deamination of [15N] glutamate to provide 2-oxoglutarate and [15N] ammonium. In the presence of cadmium, excess [15N] ammonium was incorporated into asparagine, which served as an additional detoxification molecule. In the presence both of methionine... |

This study was conducted to evaluate the effect of foliar Pi application on gas exchange and yield of bean genotypes submitted to a mild water deficit at the pre-flowering stage. In the first experiment, when extra Pi (10 g.L-1) was sprayed on leaves during water stress or during recovery, there was no effect on gas exchange or yield in the A320, Carioca and Ouro Negro genotypes. However, net CO2 assimilation (A) of A320 and Ouro Negro was less affected, but not significantly, than Carioca at the end of the stress, when Pi was supplied five days before water deficit. In the second experiment, two different doses of Pi (10 and 20 g.Pi L-1) were sprayed five days before water deficit on the Carioca genotype. During the last three days of the mild water... |
